Effects of BA and NAA Combinations and Gamma Radiation on In Vitro Culture of Philodendron ‘Cherry Red’

บทคัดย่อ

การทดลองนี้มีวัตถุประสงค์เพื่อศึกษาผลของ 6-benzyladenine (BA) ร่วมกับ naphthalene acetic acid (NAA) ต่อการเพิ่มจำนวนยอดของฟิโลเดนดรอน เชอร์รี่เรด และผลของการฉายรังสี แกมมาที่มีต่อลักษณะฟิโลเดนดรอน เชอร์รี่เรด ในสภาพปลอดเชื้อ

โดยแบ่งเป็น 2 การทดลอง ได้แก่ การทดลองที่ 1 นำชิ้นส่วนปลายยอดของฟิโลเดนดรอน เชอร์รี่เรด มาเพาะเลี้ยงบนอาหารสูตร Murashige และ Skoog (MS) ที่เติมสารควบคุมการเจริญเติบโต BA ร่วมกับ NAA จำนวน 8 สูตร การทดลองที่ 2 ต้นฟิโลเดนดรอน เชอร์รี่เรด ที่เพาะเลี้ยงบนอาหาร สูตร MS เป็นเวลา 20 สัปดาห์ ไปฉายรังสีแกมมาที่ปริมาณรังสี 0, 10, 20, 30, 40 และ 50 เกรย์ ที่สถาบันเทคโนโลยีนิวเคลียร์แห่งชาติ จังหวัดนครนายก หลังจากนั้นนำ มาเพาะเลี้ยงในห้องภายใต้อุณหภูมิ 25±2 องศาเซลเซียส ความเข้มแสง 2,000 ลักซ์ ให้แสงเป็นเวลา 16 ชั่วโมงต่อวัน ที่ห้องปฏิบัติการเพาะเลี้ยงเนื้อเยื่อพืช สำนักวิจัยพัฒนาเทคโนโลยีชีวภาพ จังหวัดปทุมธานี ทำการทดลองระหว่างเดือนมีนาคม 2560 – สิงหาคม 2561 วางแผนการทดลองแบบสุ่มสมบูรณ์ (CRD)

ผลการทดลองที่ 1 พบว่า ชิ้นส่วนปลายยอดของฟิโลเดนดรอน เชอร์รี่เรด ที่เพาะเลี้ยงบนอาหารสูตร MS ที่เติม BA 1 มิลลิกรัมต่อลิตร ร่วมกับ NAA 0.5 มิลลิกรัมต่อลิตร ให้จำนวนยอดเฉลี่ยมากที่สุด คือ 8.56 ยอด นอกจากนี้ยังส่งผลต่อความสูงยอดเฉลี่ยและจำนวนใบเฉลี่ยสูงสุดเท่ากับ 18.54 มิลลิเมตร และ 36.78 ใบ ตามลำดับ หลังการเพาะเลี้ยงเป็นเวลา 12 สัปดาห์ ผลการทดลองที่ 2 แสดงให้เห็นว่า ต้นที่ผ่านการฉายรังสีที่ปริมาณ 20 เกรย์ มีความสูงต้นและจำนวนใบเฉลี่ยมากที่สุดคือ 14.46 มิลลิเมตร และ 16.22 ใบ และพบการเปลี่ยนแปลงที่ใบยอดของต้นอ่อน โดยรังสีแกมมาที่ปริมาณ 20 และ 30 เกรย์ ใบยอดมีลักษณะใบบิดและใบด่าง ตามลำดับ


ABSTRACT

The objective of this research was to investigate the effects of 6-benzyladenine (BA) and naphthaleneacetic acid (NAA) combinations and gamma radiation on in vitro culture of Philodendron ‘Cherry Red’ in sterile conditions.

There were two experiments conducted in this research. In the first experiment, the shoot tips of Philodendron ‘Cherry Red’ were cultured on 8 media of Murashige and Skoog (MS) supplemented with BA and NAA combinations, while the second experiment was conducted with plantlets cultured on MS media for 20 weeks and then irradiated with gamma rays at doses of 0, 10, 20, 30, 40 and 50 gray at the Thailand Institute of Nuclear Technology, Nakhon Nayok Province. Subsequently, the plantlets were transferred to the cultured room and maintained there under the temperature of 25±2 degree Celsius, with light intensity of 2,000 lux for 16 hours per day in the Biotechnology Research and Development Office, Pathum Thani Province. All of the experiments were conducted by using the Completely Randomized Design (CRD) from March 2017 to August 2018.

 
The results of the first experiment showed that the shoot tips of Philodendron ‘Cherry Red’ cultured on MS media supplemented with 1 mg l [superscript -1] BA and 0.5 mg l [superscript -1] NAA gave the highest average shoot number of 8.56 shoots. In addition, this medium gave the highest average shoot height and leaf number of 18.54 mm and 36.78 leaves, respectively, after 12 weeks of culture. The second experiment showed that the irradiated plantlets with gamma rays at the dose of 20 gray gave the highest average plant height and leaf number of 14.46 mm and 16.22 leaves, respectively. The treated plantlets at the dose of 20 and 30 gray showed curl leaves and variegated leaves, respectively.
